Выводы.
1. Выявлены основные закономерности, эффекты и особенности в системах люминофор — ПАВ и люминофор — тушитель люминесценции — ПАВ в мицеллярных водных растворах анионных, катионных и неионных ПАВ, позволяющие улучшить характеристики флуоресцентного определения ПАУ и дающие возможность проводить их определение методом фосфоресценции при комнатной температуре (ФКТ) и сенсибилизированной ФКТ. Проведен и обоснован выбор доноров энергии, позволивших наблюдать перенос энергии в триплетном состоянии и сенсибилизованную фосфоресценцию ПАУ при комнатной температуре в мицеллах ДДС в присутствии внешнего тяжелого атома таллия (I).
2. Изучено влияние растворов ПАВ на эффективность взаимодействия ПАУ и тушителей флуоресценции. В качестве критерия эффективности взаимодействия использовалось эксимерообазование пирена (для моделирования реакции межу одинаковыми гидрофобными молекулами, солюбилизированными в ядре мицелл) и снижение интенсивности (тушение) флуоресценции ПАУ (для моделирования процессов между гидрофобными молекулами и ионами). Установлено, что на эффективность обоих типов бимолекулярного взаимодействия влияет концентрация и тип ПАВ, тип и концентрация тушителя, температура. При использовании статистики Пуассона показано, что и эксимерообразование, и тушение определяются распределением молекул между мицеллами.
3. Изучены условия возникновения сенсибилизированной фосфоресценции и факторы, влияющие на ее интенсивность и время жизни. Изучено влияние концентрации внешнего тяжелого атома на сенсибилизированную фосфоресценцию в мицеллярных растворах при комнатной температуре. Показано, что роль ионов таллия (I) как тяжелого атома состоит в увеличении скорости следующих процессов: интеркомбинационной конверсии из возбужденного синглетного в триплетное состояние молекул донора, что приводит к увеличению заселенности триплетного состояния и отражается в снижении интенсивности флуоресценции в присутствии ионов таллияизлучательной дезактивации триплетных состояний молекул донора и акцептора, что отражается в сокращении времени жизни триплетных состояний в присутствии ионов таллия.
Увеличение скорости этих процессов приводит к наблюдению следующих двух эффектов: появлению и росту интенсивности сигнала сенсибилизированной фосфоресценции ПАУсокращению времени жизни триплетных состояний и увеличению констант скорости затухания сенсибилизированной фосфоресценции ПАУ с ростом концентрации таллия.
4. Найдены оптимальные условия определения ПАУ методами флуоресценции, фосфоресценции и сенсибилизированной фосфоресценции в мицеллярных растворах ДЦС и проведено сравнение аналитических характеристик указанных методов. Впервые показано, что использование сенсибилизированной фосфоресценции позволяет повысить селективность определения ПАУ в их смеси без предварительного разделения. Рассчитаны коэффициенты селективности определения пирена, антрацена и бензантрацена в модельных растворах и выявлены условия реализации метода сенсибилизированной фосфоресценции для селективного определения отдельных ПАУ. Показано, например, что использование метода сенсибилизированной ФКТ позволяет проводить определение пирена в присутствии более чем трехсоткратных избытков нафталина, флуорена, фенантрена, хризена, флуорантена. Впервые разработаны методики определения ПАУ методом поляризационного флуоресцентного иммуноанализаоптимизированы условия специфического и групп-специфического определения. Показано, при разделении сложных смесей люминесцентные методы позволяют проводить селективное определение на основе различий в спектральных и энергетических (энергия триплетного уровня) свойствах молекул, иммунохимические на основе пространственного строения.
5. Разработаны экспресс-методики обнаружения и количественного определения следовых количеств токсикантов на основе сочетания концентрирования и определения в едином аналитическом цикле: концентрирование с помощью мицеллярных растворов использовано в сочетании с люминесцентным определением в фазе ПАВиммуноафинное концентрирование в сочетании с твердофазным иммуноферментным анализом. Разработанные методики использованы в анализе реальных объектов (вода, продукты питания, бензин, почва).
6. Найдены закономерности по влиянию природы и концентрации ПАВ и концентрируемых веществ на характеристики экстракционного извлечения из водных и неводных сред. Установлены оптимальные условия фазового разделения и экстракционного концентрирования. Впервые показана возможность экстракции ПАУ из органической фазы в мицеллярный раствор ДДС. Впервые проведено систематическое изучение экстракции анионными ПАВ в кислой среде из водных растворов. Показано, что мицеллярная экстракция позволяет достигать степени извлечения более 90% и может успешно сочетаться с дальнейшим люминесцентным определением.
7. Предложен подход, позволивший впервые сочетать иммуноафинное концентрирование в объеме геля, содержащего ковалентно связанные специфические антитела, с иммуноферментным определением на основе визуального детектирования. На его основе разработаны быстрые неинструментальные тест-методы для внелабораторного визуального детектирования целевых аналитов в сложных окрашенных матрицах, позволяющие определять и варьировать контрольный уровень. Показана применимость разработанного метода для определения индивидуальных микотоксинов охратоксина, А и афлатоксина В1 (контрольные уровни 10 нг/г и 5 нг/г соответственно, согласно рекомендациям ЕС) в образцах специй (красный перец, чили, кайенский перец, паприка, пили-пили, черный перец, белый перец, имбирь, мускат), а так же для одновременного определения при использовании двух детектирующих слоев. Представляется, что данный подход в перспективе может быть использован для определения индивидуальных веществ (пестициды, токсиканты и др) в сильноокрашенных матрицах, таких как кофе, какао, шоколад, фрукты и сухофрукты, вина.
