Тестировать реакции с участием фосфорила в активном центре фермента необычайно трудно в виду малого времени жизни фосфорилированных интермедиатов. Редко удается найти биохимическую систему и условия реакции, чтобы получить устойчивые фосфорилированные интермедиаты, имеющие непосредственное отношение к трансформации энергии АТФ.
