- Стерилизация технологического оборудования и сопутствующих ему коммуникаций насыщенным водяным паром широко используется при производстве аминокислот, ферментов, антибиотиков, и других продуктов микробиологического синтеза.
- Наличие «слабых точек» не только снижает надежность термической обработки оборудования, но и приводит к перерасходу пара и энергии.
- АНАЛИЗ МОНТАЖНЫХ СХЕМ И КОНСТРУКЦИЙ ФЕРМЕНТАТОРОВ С ПОЗИЦИИ СТЕРИЛИЗУЕМОСТИ.
- Монтажная схема ферментатора с нижним спуском представлена на Рис. 1. Данная схема предназначена для обеспечения асептических условий процесса ферментации.
- Отработанный воздух отводится из ферментатора по трубопроводу 6 при открытом вентиле 5 и закрытом вентиле 4. Рассматриваемый узел стерилизуется вместе с ферментатором при открытом вентиле 5 путем подачи пара из аппарата к открытому трубному окончанию.
- Подача жидких добавок производится по трубопроводу 26 (полная схема на Рис. 1 не показана), имеющему конструкцию, аналогичную трубопроводам 10 и 14.
- При отборе готовой продукции открываются вентили 27, 30 и 29, а вентили 28 и 31 закрыты. При необходимости сброса содержимого ферментатора в канализацию открывают вентили 27, 30 и 31, при этом вентили 29 и 31 закрыты.
- При рассмотрении монтажных схем аппаратов, например аппарата с нижним спуском, можно выделить ряд узлов, примеры которых приведены на соответствующих рисунках

- Таким образом, рассмотрение узлов обвязки аппаратов с точки зрения обеспечения их эффективной стерилизации позволяет выделить несколько характерных «слабых точек», стерилизация которых затруднена.
- Узлы 7. Тупиковые полости ферментаторов.
- При введении через штуцер термометра сопротивления в нем образуется тупиковая полость. При установке манометра с разделительной мембраной или при наличии заглушек на отдельных аппаратных штуцерах также образуются сложные тупиковые полости.
- Некоторые результаты экспериментального исследования режимов стерилизации аппарата вместимостью 63 м³.1.
- Таким образом, при недостаточной пропускной способности узла нижнего спуска образуется область с пониженной температурой, уменьшающей эффективность стерилизации, что приводит к загрязнению операции.152
- Оценка эффективности процесса стерилизации.
- Так как оценка режимов стерилизации оборудования и коммуникаций по своесути является вероятностной, то необходимо задаться некоторым уровнем стерильности ферментаторов.
- При этом постулизируется, что попадание в ферментатор даже одного постороннего микроорганизма ведет к загрязнению ферментатора.
- Количество оставшихся микроорганизмов может быть определено из критерия стерилизации:1. V = 1п (ЛГ0 /ЛО (2)где:

- Ы0 -количество жизнеспособных микроорганизмов в заданном объеме до тепловой обработки

- Количество оставшихся микроорганизмов может быть определено исходя из определения критерия стерилизации:1. У=1п (лул0,где:

- Из микроорганизмов, встречающихся в питательной среде и на поверхностях оборудования, особенно трудно инактивируются в тепловом поле споры рода ВасП
- Из приведенных соотношений и принятого допущения можно получить выражение для расчета показателя асептической эффективности:
